Live Cell Imaging Facility
The system comprises:
- Olympus IX70 inverted microscope (see under Light microscopy)
- Fluorescence System
- MetaFluor and MetaMorph software for fluorescence image acquisition.
- Hamamatsu multi-format cooled CCD camera
- TMC Isolation table
This system is currently located in Rm S445, Anderson Stuart Building but will be re-located to N135 in October 2009. This system currently has limited use since it is to be re-located into PC2 facilities but it is available for use on request.
The Sutter DG-4 Wavelength Switcher is a combined Xenon Arc lamp and high speed wavelength switcher, ideal for fast ion imaging where image ratios at different excitation wavelengths are required at high frame rates. It comprises:
- Built-in 175W ozone-free Xenon lamp
- Uses up to 4 standard (25mm diameter) interference filters
- Beam is switched between filters in less than 1.2 ms
- Allows video-rate ratio imaging (depending on camera hardware)
- Liquid light guide provides vibration isolation from microscope
- Integral shuttering and neutral density filtering
Filters
The system is currently configured for ratio imaging of:
- FURA-2 (Omega filter XF04: Exc1 340AF15, Exc2 380AF15, DM 415 DCLP, Em 510WB40).
- Di-8-ANEPPS (Omega filter XF17: Exc1 490DF20, Exc2 440AF21, DM 550DCLP, Em 660DF50)
Software
- Fluorescence image acquisition is controlled by MetaFluor (Universal Imaging)
- Image processing is achieved with MetaMorph (Universal Imaging)
